چکیده

This paper reports a study of the adsorption of four proteins-fibrinogen, lysozyme, pyruvate kinase, and RNAse A-to self-assembled monolayers (SAMs) on gold. The SAMs examined were derived from thiols of the structure HS(CH2)loR, where R was CH3, CHzOH, and oligo(ethy1eneoxide). Monolayers that contained a sufficiently large mole fraction of alkanethiolate groups terminated in oligo(ethy1eneoxide) chains resisted the kinetically irreversible, nonspecific adsorption of all four proteins. Longer chains of oligo(ethy1ene oxide) were resistant at lower mole fractions in the monolayer. Resistance to the adsorption of proteins increased with the length of the oligo(ethy1ene oxide) chain: the smallest mole fraction of chains that prevented adsorption was proportional to where n represents the number of ethylene oxide units per chain. Termination of the oligo(ethy1ene oxide) chains with a methoxy group instead of a hydroxyl group had little or no effect on the amount of protein adsorbed. The amount of pyruvate kinase that adsorbed to mixed SAMs containing hexa(ethy1ene oxide)-terminated chains depended upon the temperature. When the mole fraction of oligo(ethy1ene oxide) groups in the monolayer was below the level needed to prevent adsorption, more pyruvate kinase adsorbed to the monolayer at 37 "C than at 25 "C. No difference was observed between adsorption at 25 and 4 "C.

Protein adsorption to oligo(ethylene glycol) self-assembled monolayers: experiments with fibrinogen, heparinized plasma, and serum.

Low protein adsorption is believed advantageous for blood-contacting materials and ethylene glycols (EG)-based polymeric compounds are often attached to surfaces for this purpose. In the present study, the adsorption of fibrinogen, serum, and plasma were studied by ellipsometry on a series of well-defined oligo(EG) terminated alkane-thiols self-assembled on gold. The layers were prepared with c...

Physical properties of self-assembled monolayers of Mercapto Oligo (ethylene oxide) methyl ether on gold.

Physical properties (chemical adsorption rate, viscoelasticity, thickness, and conformation) for self-assembled monolayers (SAMs) of mercapto oligo (ethylene oxide) methyl ethers on gold were determined by quartz crystal microbalance (QCM). The values of thickness, shear viscosity and elastic shear modulus of SAMs increase with unit number of oligo (ethylene oxide) segment. However, the chemica...

Formation of tetra(ethylene oxide) terminated Si-C linked monolayers and their derivatization with glycine: an example of a generic strategy for the immobilization of biomolecules on silicon.

Surface modification with oligo(ethylene oxide) functionalized monolayers terminated with reactive headgroups constitutes a powerful strategy to provide specific coupling of biomolecules with simultaneous protection from nonspecific adsorption on surfaces for the preparation of biorecognition interfaces. To date, oligo(ethylene oxide) functionalized monolayer-forming molecules which can be acti...

Self-assembled organic monolayers: model systems for studying adsorption of proteins at surfaces.

Self-assembled monolayers (SAMs) of omega-functionalized long-chain alkanethiolates on gold films are excellent model systems with which to study the interactions of proteins with organic surfaces. Monolayers containing mixtures of hydrophobic (methyl-terminated) and hydrophilic [hydroxyl-, maltose-, and hexa(ethylene glycol)-terminated] alkanethiols can be tailored to select specific degrees o...

One step growth of protein antifouling surfaces: monolayers of poly(ethylene oxide) (PEO) derivatives on oxidized and hydrogen-passivated silicon surfaces.

We compare two routes for creating protein adsorption-resistant self-assembled monolayers (SAMs) by chemical modification of silicon surfaces with poly(ethylene oxide) (PEO) oligomeric derivatives. The first route involves the assembly of 2-methyl[(polyethyleneoxy)propyl]trichlorosilane (Cl3SiMPEO) films onto oxidized silicon surfaces (OH-SiO(x)) either by a liquid-phase process at room tempera...
